Flotillin-2
Flotillin-2 (FLOT2) is a membrane-associated scaffolding protein that participates in caveolar membrane organization and the formation of caveolae-like vesicles (UniProt: Q14254). It may also contribute to epidermal cell adhesion and epidermal structure. The protein localizes to specific subcellular compartments and is 428 amino acids in length.
While UniProt contains no curated disease associations for FLOT2, the protein has been identified as relevant to Alzheimer's Disease pathology through quantitative proteomics. In post-mortem AD brain tissue compared to age-matched controls, FLOT2 is consistently downregulated across subcellular fractions (mean log2 fold-change: −0.61) (Chaparral AD proteomics). This reduction was measured using TMT-labeled tandem mass spectrometry across four subcellular compartments (P2, P3, S2, S3), indicating a reliable downregulation signal in AD neuropathology.
The functional significance of FLOT2 reduction in AD remains to be elucidated, though impaired caveolar organization and membrane trafficking could potentially contribute to neuronal dysfunction or accumulation of pathogenic proteins.
