Sorting nexin-32
Sorting nexin-32 (SNX32) is a BAR domain-containing protein of 403 amino acids that functions in endosomal trafficking and cargo sorting. It mediates transport of multiple receptor proteins including transferrin receptor (TFRC), the cation-independent mannose-6-phosphate receptor (IGF2R), and epidermal growth factor receptor (EGFR) from endosomes to the trans-Golgi network or lysosomes for degradation (UniProt: Q86XE0). SNX32 also plays a role in neurite outgrowth through regulation of basigin trafficking to the cell surface.
The protein operates in endosomal and trafficking pathways critical for receptor homeostasis and cellular signaling. It functions redundantly with other sorting nexins, particularly SNX5, in coordinating EGFR degradation and IGF2R recycling. No disease associations are annotated in the UniProt record.
SNX32 is downregulated in Alzheimer's disease brain tissue (Chaparral AD proteomics). Human post-mortem AD brain samples show a mean log2 fold-change of −0.71 compared to age-matched controls in TMT-labeled proteomics across subcellular fractions. This reduced expression may impact endosomal trafficking and receptor turnover pathways relevant to neuronal dysfunction in AD.
