Sorting nexin-2
Sorting nexin-2 (SNX2) is a membrane-trafficking protein that binds phosphoinositide-enriched membranes and functions as a component of the retromer SNX-BAR subcomplex (UniProt: O60749). It mediates retrograde transport of cargo proteins from endosomes to the trans-Golgi network and promotes endosome-to-plasma membrane recycling through membrane deformation and tubule formation. SNX2 also supports KALRN- and RHOG-dependent membrane remodeling independent of retromer function.
SNX2 participates in intracellular trafficking pathways critical for cellular protein homeostasis and vesicular transport across multiple compartments. The protein interacts with key trafficking regulators and possesses intrinsic membrane-remodeling activity. No UniProt-annotated disease associations are listed.
SNX2 is upregulated in Alzheimer's disease brain tissue relative to age-matched controls (mean log2 fold-change +0.64; Chaparral AD proteomics). This upregulation was detected in post-mortem human AD brain via TMT-labeled mass spectrometry across subcellular fractions. The elevation in SNX2 may reflect compensatory activation of endosomal-recycling pathways or alterations in membrane trafficking associated with neurodegeneration.
