Microtubule-associated protein RP/EB family member 1
MAPRE1 (microtubule-associated protein RP/EB family member 1) is a plus-end tracking protein (+TIP) that binds to the plus-end of microtubules and regulates microtubule cytoskeleton dynamics (UniProt: Q15691). It recruits other +TIP proteins by recognizing conserved Ser-X-Leu-Pro motifs and promotes microtubule nucleation, elongation, and mitotic spindle positioning. MAPRE1 also regulates minus-end microtubule organization at the Golgi apparatus through interaction with CAMSAP2, and facilitates autophagosome transport.
MAPRE1 is involved in multiple cellular processes including chromosome segregation, spindle checkpoint function, and polarized cell migration (UniProt: Q15691). The UniProt record does not list inherent disease associations.
In Alzheimer's disease, MAPRE1 is upregulated in post-mortem human brain tissue compared to age-matched controls (Chaparral AD proteomics). The protein showed elevated levels with a mean log2 fold-change of 0.25 across subcellular fractions in a TMT-labeled proteomics study. This upregulation may reflect compensatory responses to microtubule destabilization or neuronal cytoskeletal stress characteristic of AD pathology.
