PDZ and LIM domain protein 7 (PDLIM7) is a 457-amino acid scaffold protein that facilitates coordinated assembly and localization of proteins through its PDZ domain, anchoring LIM-binding proteins to actin filaments in skeletal muscle and nonmuscle tissues (UniProt: Q9NR12). The protein is involved in bone formation via both direct and endochondral mechanisms, as well as fracture repair, and participates in BMP6 signaling pathways.
PDLIM7 is broadly expressed across tissues involved in structural organization and development. No primary disease associations are documented in UniProt curated data; however, emerging evidence suggests relevance to neurodegenerative processes through its actin-regulatory functions.
PDLIM7 is upregulated in Alzheimer's Disease brain tissue. Chaparral AD proteomics data from post-mortem human AD brain compared to age-matched controls shows PDLIM7 is elevated with a mean log2 fold-change of 0.86 across subcellular fractions (P2, P3, S2, S3) in TMT-labeled quantitative proteomics analysis. This upregulation may reflect compensatory cytoskeletal remodeling or pathological protein aggregation responses characteristic of AD neurodegeneration.
