EH domain-containing protein 4
EHD4 (EH domain-containing protein 4) is an ATP- and membrane-binding protein that regulates membrane reorganization and tubulation through ATP hydrolysis. It participates in early endosomal transport and, in complex with PACSIN2 and MICALL1, controls CDH5 trafficking at asymmetric adherens junctions during sprouting angiogenesis by forming recycling endosome-like tubular structures (UniProt: Q9H223).
The protein functions in endosomal and membrane trafficking pathways with relevance to cell adhesion and vascular biology. No disease associations are documented in the UniProt record.
EHD4 shows increased abundance in Alzheimer's Disease brain tissue relative to age-matched controls (Chaparral AD proteomics). Analysis of post-mortem human AD brain versus controls using TMT-labeled quantitative proteomics across four subcellular fractions (P2, P3, S2, S3) detected EHD4 upregulation with a mean log2 fold-change of 0.58, suggesting a consistent elevation across at least two fractions.
