Pericentriolar material 1 protein
PCM1 (pericentriolar material 1 protein) is a 2024-amino acid scaffolding protein essential for centrosome assembly and microtubule anchoring (UniProt: Q15154). It localizes centrosomal proteins including CEP250, CETN3, and PCNT, and is also required for cilium biogenesis through recruitment of the BBSome complex to centriolar satellites. PCM1 additionally recruits the tubulin polyglutamylase complex to these structures.
PCM1 functions in centrosomal and ciliary biology across tissues expressing centrosomes and cilia. While no disease associations are listed in UniProt, the protein's role in microtubule organization and centrosomal integrity suggests potential relevance to neurodegenerative pathways affecting neuronal structure.
PCM1 is down-regulated in Alzheimer's Disease brain tissue. Analysis of post-mortem AD brain versus age-matched controls by TMT-labeled proteomics detected PCM1 with a mean log2 fold-change of −0.33 across subcellular fractions (Chaparral AD proteomics). This modest reduction may reflect alterations in centrosomal function or microtubule dynamics in the AD brain.
