Myosin-10 (MYH10) is a molecular motor protein belonging to the unconventional myosin family, involved in actin-based cytoskeletal dynamics and cellular force generation (UniProt: P35580-3). The protein plays roles in cell migration, morphogenesis, and membrane trafficking through its interaction with actin filaments. Isoform 3 represents one of several alternatively spliced variants of this 231 kDa protein.

In Alzheimer's disease, isoform 3 of myosin-10 is downregulated in post-mortem AD brain tissue compared to age-matched controls (Chaparral AD proteomics). Analysis of human post-mortem AD brain using TMT-labeled quantitative proteomics across four subcellular fractions (P2, P3, S2, S3) showed a mean log2 fold-change of −0.456, indicating modest but consistent reduction of this isoform in the AD brain proteome. The functional significance of this downregulation in AD pathogenesis remains to be elucidated.
