Vinexin
Vinexin (SORBS3) is a cytoplasmic adapter protein that regulates actin dynamics and cell signaling. The alpha isoform promotes actin stress fiber formation, while the beta isoform facilitates cell spreading and enhances JNK/SAPK activation in response to EGF stimulation through its third SH3 domain (UniProt: O60504).
Vinexin is broadly expressed and functions as a scaffolding protein linking receptor tyrosine kinases to the actin cytoskeleton, supporting cell migration and morphogenesis. No disease associations are annotated in UniProt.
Vinexin is upregulated in Alzheimer's disease brain tissue. Analysis of post-mortem AD brain compared to age-matched controls using TMT-labeled proteomics across four subcellular fractions revealed increased vinexin abundance with a mean log2 fold-change of +0.54 (Chaparral AD proteomics). This upregulation may reflect altered cytoskeletal remodeling or compensatory responses to neuronal stress in AD pathology.
