WD repeat-containing protein 37
WDR37 is a WD repeat-containing protein of 494 amino acids that plays a role in endoplasmic reticulum calcium handling in lymphocytes. Together with PACS1, it is required for stabilizing peripheral lymphocyte populations (UniProt: Q9Y2I8). The protein is implicated in neurooculocardiogenitourinary syndrome (NOCGUS), a rare autosomal dominant multisystem disorder featuring severe neurological dysfunction, structural brain abnormalities, seizures, and multiple organ involvement (UniProt: Q9Y2I8).
WDR37 is downregulated in Alzheimer's disease brain tissue. Chaparral AD proteomics analysis of post-mortem AD brain versus age-matched controls using TMT-labeled quantitative mass spectrometry across four subcellular fractions detected reduced WDR37 abundance with a mean log2 fold-change of −0.43 (Chaparral AD proteomics). This downregulation was observed in one fraction from the proteomic dataset, suggesting potential dysregulation of calcium signaling or lymphocyte-related immune processes in the AD brain microenvironment.
