E3 ubiquitin-protein ligase NRDP1
NRDP1 (encoded by RNF41) is an E3 ubiquitin-protein ligase that regulates protein degradation through polyubiquitination of multiple substrates including MYD88, ERBB3, and BIRC6 (UniProt: Q9H4P4). It functions in immune signaling by negatively regulating MYD88-dependent cytokine production and promoting type I interferon responses, as well as in mitochondrial quality control through the RNF41-PRKN pathway that regulates autophagosome-lysosome fusion during mitophagy (UniProt: Q9H4P4).
NRDP1 is involved in hematopoietic progenitor cell differentiation and apoptotic regulation. No UniProt-curated disease associations are currently listed for this protein (UniProt: Q9H4P4).
In Alzheimer's Disease, NRDP1 shows reduced protein abundance in post-mortem AD brain tissue compared to age-matched controls, with a mean log2 fold-change of −0.31 (Chaparral AD proteomics). This down-regulation was detected across a single subcellular fraction in TMT-labeled proteomics analysis of four fractions from AD brain tissue, suggesting potential impairment of proteolytic and mitophagy pathways in AD pathology.
