Protein kinase C epsilon type
Protein kinase C epsilon type (PRKCE) is a calcium-independent, phospholipid- and diacylglycerol-dependent serine/threonine kinase that regulates multiple cellular processes including cytoskeletal dynamics, cell adhesion, migration, and cell cycle progression (UniProt: Q02156). In neurons, it promotes growth and modulates ion channel activity; it also mediates synaptic potentiation and regulates GABA receptor and heat-activated channel function. The enzyme phosphorylates numerous substrates including structural proteins (MARCKS, vimentin, keratin-8) and signaling molecules (PTK2/FAK, STAT3, TICAM2).
PRKCE plays roles in cardiac myocyte physiology, immune responses via TLR4 signaling, and apoptosis regulation across multiple tissues (UniProt: Q02156). No UniProt-curated disease associations are listed for the protein itself.
PRKCE is downregulated in Alzheimer's disease brain tissue. Human post-mortem AD brain compared to age-matched controls showed reduced PRKCE abundance with a mean log2 fold-change of −0.54 across subcellular fractions (P2, P3, S2, S3) detected by quantitative mass spectrometry (Chaparral AD proteomics). This reduction may be relevant to AD-associated neuronal dysfunction, though the functional consequences remain to be determined.
