Pre-mRNA-splicing factor SPF27
SPF27 (encoded by BCAS2) is a 225-amino acid protein essential for pre-mRNA splicing as a core component of the activated spliceosome (UniProt: O75934). It functions as part of the PRP19-CDC5L complex, which forms an integral part of the spliceosome and is required for activating pre-mRNA splicing. The complex may also play a role in DNA damage response pathways.
SPF27 is widely expressed and functions in fundamental gene expression processes through its role in spliceosome assembly and activation. The protein appears to have a scaffolding function, contacting all other core components of the spliceosome complex (UniProt: O75934). No disease associations are reported in UniProt.
SPF27 shows reduced abundance in Alzheimer's Disease brain tissue. In post-mortem AD brain compared to age-matched controls, SPF27 was significantly down-regulated with a mean log2 fold-change of −1.37 across TMT-labeled proteomics analysis of subcellular fractions (Chaparral AD proteomics). This downregulation suggests potential involvement of impaired splicing activity in AD pathophysiology, though the functional consequences remain to be determined.
